Bien qu'il s'agisse d'une suite à To-Love-Ru, nous y retrouvons tous nos personnages principaux, mais avec quelques ajouts. Rito continue d'être pris dans des scénarios ecchi haut de gamme. Les soeurs alien Deviluke utilisent toujours leurs capacités uniques pour faire avouer son amour à Rito. Nana et Momo sont transférées dans l'école et rencontrent Mea, qui semble être une fille insouciante et joyeuse. Momo de son côté veut partager Rito et créer un harem pour lui. Alors qu'elle prépare son plan, elle se confronte à l'Ombre Dorée, également connue sous le nom de Yami (Le nom japonais est Konjiki no Yami). Ils sont interrompus par des étudiants possédés qui commencent à se battre avec une force anormale. Un nouveau complot apparaît sous la forme du personnage mystérieux qui contrôle les élèves, ceci afin de tester les compétences en assassinat de Yami. Qui est ce nouvel ennemi ?
